Western blot analysis of GRK6 expression in HEK293T (A), HepG2 (B), H9C2 (C), rat liver (D), mouse heart (E) whole cell lysates. (Predicted band size: 65 kD; Observed band size: 80 kD)
Immunohistochemical analysis of GRK6 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of GRK6 staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Function	Specifically phosphorylates the activated forms of G protein- coupled receptors (PubMed:19423652, PubMed:20048153). Such receptor phosphorylation initiates beta-arrestin-mediated receptor internalization, and signaling events leading to their desensitization (PubMed:19423652, PubMed:20048153). Seems to be involved in the desensitization of D2-like dopamine receptors in striatum and chemokine receptor CXCR4 which is critical for CXCL12-induced cell chemotaxis (PubMed:20048153). Phosphorylates rhodopsin (RHO) (in vitro) and a non G protein-coupled receptor: LRP6 during Wnt signaling (in vitro) (PubMed:19801552).
