Western blot analysis of ABCD3 expression in HepG2 (A) whole cell lysates. (Predicted band size: 75 kD; Observed band size: 70 kD)
Immunofluorescent analysis of ABCD3 staining in HepG2 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with an AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AF594 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).

Function	ATP-dependent transporter of the ATP-binding cassette (ABC) family that catalyzes the transport of long- and very-long-chain fatty acyl-CoAs and probably bile acyl-CoAs from the cytosol to the peroxisome matrix for beta-oxydation. Appears to be specifically involved in the peroxisomal transport of branched-chain fatty acyl-CoAs (BCFA-CoAs) and dicarboxylic acyl CoAs (DCA-CoAs) (PubMed:11248239, PubMed:23671276, PubMed:24333844, PubMed:25168382, PubMed:29397936, PubMed:39223112, PubMed:41428872). Has fatty acyl-CoA thioesterase and ATPase activities. Probably hydrolyzes fatty acyl-CoAs into free fatty acids prior to their ATP-dependent transport into peroxisomes.
